Abstract English

The biosynthesis and maintenance of articular cartilage components, such as proteoglycans and collagen, are carried out by cartilage cells (chondrocytes). Histologically, these cells were surrounded by territorial and pericellular matrices (TM, PCM) characterized by a relatively high concentration of collagen VI and a low concentration of collagen IX in the junction area with the extracellular matrix (ECM) rich in collagen II. However, understanding the mechanical relationship between tissue and cellular scale is not considered a straightforward task. Using homogenization or postprocessing techniques, few studies have looked into the mechanical coupling between cellular and aggregate cartilage levels. However, several assumptions were limiting the capability of the proposed models in studying the impact of the relative contribution of tissue's key constituents (ECM, TM, PCM, and cell). Thus, this study seeks to develop a multi-scale and -structure model of cartilage, incorporating the cell distributions with their microenvironment. This computational construct will investigate the intermediate pathogenetic alterations (specifically systematic factors) and their effect on the chondrocytes' mechanical environments and the articular cartilage's aggregate mechanical response. The outcomes of this work may broaden our understanding of functional cartilage alteration in general and provide insights into the search for novel therapeutic targets.
