Abstract Arabic
(Multiplex assays).
Abstract English
Human obesity/type-2 diabetes (T2D) prevalence is escalating globally, and in Kuwait as well. A positive energy balance due to high-caloric food-intake with sedentary lifestyle result in storage of superfluous energy as triglycerides in the white adipose tissue. The white adipose tissue mainly comprises of adipocytes and immune cells. In obesity, immunometabolic changes in white adipose tissue disrupt the homeostasis and lead to chronic low-grade inflammation called metabolic inflammation. Notably, enhanced metabolic activity/energy demand in the expanding adipose tissue require neovascularization over time to compensate needs for additional oxygen and nutrients. During this time, a state of oxidative stress is established within the adipose microenvironment. Our preliminary data show increased expression of TLRs-2/4/8/10 and stress regulator IRF5 in human adipose tissue in obesity/T2D; changes correlating with BMI. We, therefore, hypothesize that oxidative stress can induce/promote TLR4/IRF5 expression in macrophages and adipocytes – two most critical players in the fat. In this study, human/mouse cells will be subjected to oxidative stress by H2O2 and/or 3%O2 hypoxia to induce reactive oxygen species (ROS). ROS induction will be assessed by DCFH-DA staining and super oxide dismutase (SOD) activity while cytotoxic effects will be determined using dye-exclusion and/or MTT assays. Expression changes in TLR4/IRF5 will be measured at both mRNA and protein levels using real-time RT-PCR and flow cytometry (or confocal microscopy, western blotting), respectively. Multiplex cytokines/chemokines assays will be used to measure secretory responses. TLR4/IRF5 siRNAs will be transfected for target gene suppression in macrophages/adipocytes while pathways’ inhibitors will be used to identify the signaling mechanism(s) involved. Effects on metabolic function will be assessed using glucose uptake assays. Data (mean±SEM) will be analyzed using t-test, ANOVA, Pearson’s correlation, and multivariate analyses as appropriate. The information gained will fill gaps in knowledge, help identify target(s) of predictive/therapeutic significance and pave way for in vivo model studies.
