على الرغم من ارتفاع معدل الإصابة بداء السكري من النوع 2 (T2D) والسمنة في شبه الجزيرة العربية، إلا أنه تم إجراء عدد قليل من الدراسات الوراثية على مستوى الجينوم لتحديد المتغيرات المرتبطة بمخاطر السكري (DM) ولم يتم تطوير أدوات لتقييم المخاطر الجينية للسكان في المنطقة. في السابق، قمنا بإجراء فحص جيني على مستوى الجينوم وتسلسل كامل للأكواد الجينية (الـ Whole-exome sequencing) على عينة صغيرة من السكان العرب من الكويت؛ كما قمنا بإجراء تحليلات جينومية ووراثية. على الرغم من أن العمل أسفر عن عدد كبير من المنشورات حول جينوم السكان، إلا أن التقدم كان محدودًا في تحديد المتغيرات الوراثية المرتبطة بمخاطر السكري. في الواقع، لم ينجح أي من العلامات التي تم اختبارها في اجتياز إحصائيات الدلالة الجينومية، وكان السبب الرئيسي في ذلك هو أن الدراسة كانت ذات طاقة إحصائية ضعيفة. وبالتالي، هناك حاجة إلى مواصلة هذه المبادرة من خلال تطبيق أساليب بديلة ومتقدمة على البيانات المستخلصة من دراسات GWAS (دراسات الارتباط على مستوى الجينوم) والـ Exome من أجل فك تشفير البنية الوراثية للسكري في الكويت. ومن ثم، نعتزم استخدام النتائج المستخلصة حول البنية الوراثية للسكري في السكان الكويتيين لتطوير أدوات لتقييم المخاطر التي ستساهم بشكل كبير في تعزيز جهود الوقاية من السكري.
Abstract English
Despite a high incidence of T2D and obesity in the Arabian Peninsula, few genome-wide genetic studies on risk variants for DM have been performed and no genetic risk assessment tools have been developed for populations from the region. Previously, we performed genome-wide genotyping and whole-exome sequencing on a small-sized Arab cohort from Kuwait; we further performed genomics and genetics analysis. Although the work resulted in a considerable number of publications on population genomics, only limited progress was made in identifying genetic risk variants for DM. Virtually none of the tested markers passed the statistics at genome-wide significance, the main reason being that the study was underpowered. Thus, there is a need to further pursue this initiative by applying alternative and advanced approaches on the generated GWAS and exome data in order to decipher the genetic architecture of DM in Kuwait. Subsequently, we intend to use the derived results on the genetic architecture of DM in the Kuwaiti population to develop risk assessment tools that will greatly enhance DM prevention efforts.
