يهدف البحث إلى دراسة قدرة مختلف المولدات المضادة الموجودة في جراثيم السل على تنشيط الخلايا التائية والتي قد تؤدي إلى تحفيز مناعة واقية (Th1) أو مناعة باثولوجية (Th2) في كل من مرضى السل والأصحاء. وسيتم ذلك بفحص عدد من المولدات المضادة المنقاة وجمعها بعد زراعة البكتريا عن طريق الموجات الصوتية ومجمل الخلايا البكتيرية ثم تحلل تكاثر الخلايا التائية من مرضى السل غير المعالجين، المعالجين والأصحاء المطعمين. ثم سيتم اختيار مولدات المضادات القادرة على التمييز بين المرضى والأصحاء أو المولدات القادرة على استثارة الخلايا التائية الواقية لإجراء المزيد من الدراسات التفصيلية. ثم ستفحص الخلايا التائية لمدى استثارتها وافرازها السيتوكينات بوجود آكلة الخلايا الكبيرة. للتعرف على طبيعة الخلايا وخصائصها وقدرتها على إفراز السيتوكينات على مستوى الجينات وتحديد نوعية السيتوكينات المفرزة، وقدرتها على تثبيط نمو بكتريا السل داخل آكلة الخلايا الكبيرة البشرية. ويمكن القول أن تحديد الخواص المناعية لمختلف مولدات المضادات على التعرف على المولدات المهمة في التشخيص المبكر والدقيق وفي تطوير تطعيم واقٍ أفضل يتم انتاجه بكميات كبيرة وبتكلفة أقل مع تجنب استعمال البكتريا الجرثومية وذلك باستخدام صناعة الببتيدات.

Abstract English

The project proposes to test various antigens and epitopes of M. tuberculosis for their ability to induce T cells of protective (Th1) or pathogenic (Th2) phenotypes in healthy subjects and TB patients. To identify the type of T cell responses mounted (Th1 and Th2) to various mycobacerial antigens, a set of purified antigens of M. tuberculosis will be tested. The antigens will include the defined secreted antigens, i.e. ESAT6, MPT59, MPT64, MPB70, MPT83; non-secreted antigen, i.e. 10kDa, 38kDa, 65 kDa, 70 kDa; fractionated secreted antigens, culture filtrates containing all the secreted antigens, cell wall, sonicate and whole mycobacteria. Antigen-induced proliferation and IFN- secretion will be studied using peripheral blood mononuclear cells (PBMC) from untreated and cured TB patients, BCG vaccinated healthy donors and PPD positive healthy contacts of TB patients with varied MHC background. T cells lines from PBMC of selected healthy donors and TB patients expressing different HLA-DR molecules will be established against the defined antigens and then tested with homologous antigens and their synthetic peptides covering the entire amino acid sequence of each antigen. The antigens and peptide epitopes recognised by T cells in association with multiple HLA-DR molecules will be preferred in diagnosis and vaccine development. To identify such antigens and epitopes, the T cell lines will be tested for proliferation and cytokine production in the presence of HLA-DR typed allogeneic antigen presenting cells. Furthermore, T cell clones will be established from the T cell lines to study the characteristics of T cells activated by individual epitopes at single cell level. The T clones will be tested for phenotype and antigen-peptide induced proliferation, cytotoxic activity, cytokine gene activation and cytokine secretion for Th1 (IL-2, IL-12 and IFN-) and Th2 cytokines (IL-4, IL-5 and IL-10). The protective efficacy of T cell lines and clones will be evaluated in vitro testitheir ability to inhibit bacterial growth in human macrophages pulsed with live M. tuberculosis.
