دراسة الطحالب الموجودة في المياه الاقليمية الكويتية وتنوعها كانت المهمة الأولية لمشروعنا البحثي، والتي سوف تشكل الأساس لاستغلال هذه الكائنات في انتاج الوقود الحيوي وغيره من المنتجات الثانوية القيمة (مثل المركبات النشطة بيولوجيا، والغذاء، والأعلاف). لقد كان من الضروري اختيار الطحالب الدقيقة المعزولة محليا من المياه الساحلية الكويتية التي تستطيع انتاج كميات ضخمة من الدهون. أثناء المرحلة الأولية ( فبراير 2016 - فبراير 2017)، تم تجميع عدد من السلالات الأصلية المحلية من نقاط ساحلية مختلفة. تم عزل ما مجموعه 108 سلالة تنتمي لعوائل باسيلاريوفيسي، كلوروفيسي و سيانوفيسي وذلك باستخدام الأوساط الغذائية الغنية بمكوناتها والخاصة بتنمية أكبر عدد من الطحالب الدقيقة، تلاها عزل مجموعة واسعة من الطحالب الدقيق أحادية النسل. وسيكون من الممكن إجراء دراسات مقارنة مستقبلا لدراسة تواجد هذه الأنواع من الطحالب الدقيقة وصفاتها الحيوية بمساعدة هذه المجموعة. حتى الآن، تم الحصول على ما يقرب من اثنين وعشرين نوعا، تمثل أجناس كلوريلا، سينديسموس، نانوكلوريس، نانوكلوروبسيس، تيتراسلميس، سينكوسيستيس، ستانيرا، بليوروكابسا، سيندرا، دياتوما، غومفونيما، نيتسشيا، و نافيكولا. سوف تساهم هذه المجموعة في إنشاء قاعدة بيانات المجموعات الطحلبية الدقيقة في الكويت. تم تدريب اثنتين من طلبة مشاريع الابحاث وطالبة ماجستير على استخدام تقنيات زراعة وعزل الطحالب أحادية النوع. وعلاوة على ذلك، سيتم تدريب طالب متقدم لنيل درجة الدكتوراه على العديد من التقنيات خلال المراحل القادمة من المشروع. لقد تم لاحقا تحديد نسبة الدهون الموجودة في الطحالب المعزولة ، وذلك شأنه أن يساعد على تحديد سلالات الطحالب الدقيقة الأمثل لإنتاج كميات كبيرة من الدهون. بعد ذلك، تمت عملية التحويل الأستيري لهذه الدهون إلى استراتها من الأحماض الدهنية الميثيلية والتي حللت لاحقا باستخدام تقنية الغاز كروماتوغرافي. مجموعات الاحماض الدهنية النموذجية التي توجد عادة في وقود الديزل الحيوي الطحلبي (بما في ذلك حمض البالمتيك (16: 0) وحمض دهني (18: 0) وحمض الأوليك (18: 1) وحمض اللينوليك (18: 2) وحمض اللينولينيك (18: 3) ) وجدت في الطحالب الدقيقة المختارة، والتي سيتم اجراء المزيد من الدراسة عليها.

Abstract English

"Studying the diversity of Kuwait’s microalgae was the preliminary task of our research, and it will form the basis to
exploit these organisms in biofuel and other valuable sub-products (bioactive compounds, food, feed, etc.) production.
It was crucial to select the right lipid, hyper-producing indigenous microalgae from Kuwaiti coastal waters. During the
initial phase (February 2016 - February 2017), collection of autochthonous microalgal strains from different sampling
points was carried out. A total of 108 microalgal strains belonging to Bacillariophyceae, Chlorophyceae and Cyanophyceae groups were isolated, using a pre-enriched culturing technique, followed by the isolation of a wide range of
monospecific, monoclonal microalgae. Future comparative studies of species occurrence and biochemical features
would be feasible with the help of this collection. So far, nearly twenty two monoalgal cultures were obtained, representing Chlorella sp., Scenedesmus sp., Nannochloris sp., Nannochloropsis sp., Tetraselmis sp., Synechocystis sp.,
S. nigrescens, Staniera sp., Pleurocapsa sp., Synedra sp., Diatoma sp., Gomphonema sp., Nitzschia sp., and Navicula
sp.. These monoalgal cultures will help in the establishment of Kuwait’s Microalgae Culture Collection. Two research
project students and one MSc student were trained to use monoalgal/monoclonal culturing techniques prepare monoclonal cultures. Moreover, PhD student will be trained during the coming phases of the project. The lipid weight percentage of the isolated microalgae was gravimetrically determined, and that would permit the identification of optimal
microalgal strains as lipid hyper-producer. Algal lipids were transesterified into their fatty acid methyl esters (FAMEs)
and the FAMEs profile analysed with Gas Chromatography-Mass Spectrophotometer. Typical fatty acid groups commonly found in microalgal biodiesel (including palmitic acid (16:0), stearic acid (18:0), oleic acid (18:1), linoleic acid
(18:2), linolenic acid (18:3)) were found in the FAMEs profile of the selected microalgae, which will be selected as a
promising candidates for further study."
